Experimental studies on the carcinogenic effects of anticancer chemotherapeutics and immunosuppressive agents

1. 16 chemotherapeutics of common use from the groups of alkylants, antimetabolites, antibiotics and substances of natural origin were tested on more than 1200 rats of BR 46 strain at equitoxic dosages for their carcinogenic effects. In one experimental schedule 7% of DL50 were administered i.v. once weekly for 52 weeks. in another 3 compounds were administered at 17% of DL50, 5 times at 2 weeks intervals. The doses applied corresponded, in relation to kg of body weight, to the doses used in human therapy. 2. Distinctive carcinogenic effects were observed after application of the following alkylating substances: Dichloren (N-mustard), Mitomen (N-oxide-mustard), Endoxan (cyclophosphamide). Thio-Tepa, Trenimon, (triaziquone), Natulan (procarbazine) and Mitomvein C. The studied antimetabolites and natural products had no carcinogenic effect. 3. Whole-body X-ray irradiation at equitoxic dosage showed the same carcinogenic potency as did alkylants. 4. The carcinogenic effect of Cyclophosphamide was demonstrated not only in rats but also in mice. 5. The results are discussed in connection with the therapeutic use of the substances in anticancer chemotherapy and in immunosuppressive therapy. It is demanded that substances with cancerogenic potency should not be applied unless for vital necessity.
