The toxic effect of intramuscularly administered chlorpyrifos on different regions of rat brain was determined. The potential of chlorpyrifos to generate oxidative stress was assessed by measuring lipid peroxidation and ratio of reduced to oxidised glutathione. Male rats were treated with chlorpyrifos intramuscularly in thigh muscles for 3 days at 50, 100 and 200-mg/kg body weight. Treatment with chlorpyrifos caused significant inhibition of acetylcholinesterase activity in different regions of the brain. Oxidative stress was evident due to increase in thiobarbituric acid-reactive substances and a decrease in the ratio of reduced to oxidized glutathione in all 3 regions of the brain. The malondialdehyde level was increased significantly in all regions of the brain and the increase was dose dependent. The mid-brain had the highest level of lipid peroxidation.
