The immediate effect of neuroleptics is a dopamine receptor blocking effect which may be demonstrated as antagonism to dopamine agonists such as methylphenidate, amphetamine, and apomorphine. This receptor blocking effect is normally well correlated to the potency of the compounds against psychosis. However, in recent years it has been shown that when the receptor blockade has subsided, it is replaced by hyperreaction to the dopamine agonists. This effect is seen after both single (3,4,5,18) and repeated (6,7,15,16,19,24,25) administration and can be demonstrated both during ongoing repeated administration and after withdrawal of treatment (20,23,26).
