Fluid volume changes associated with withdrawal and restoration of steroid therapy in adrenalectomized dogs

Sodium thiocyanate, 4-aminoantipyrine, and T-1824 were used simultaneously to estimate extracellular water, total body water, and plasma volumes, respectively, in adrenalectomized Beagle dogs. While the dogs were maintained with hydrocortisone and deoxycorticosterone, the fluid compartments were measured. Therapy was discontinued and the dogs were allowed to develop mild adrenal failure, whereupon fluid compartment volumes were again determined. Hydrocortisone and deoxycorticosterone were then injected, feed and water were withheld for 24 hours, and fluid compartment quantitation was repeated. The results obtained were consistent with previous assumptions that water moves intracellularly during adrenal insufficiency. Normal fluid distribution occurred when therapy was reinstituted in the absence of food and water intake.
