Effects of estradiol on sodium and potassium balances in adrenalectomized dogs

The effects of oestradiol on Na and K balances were studied in 6 adrenalectomized dogs. Five dogs, maintained with deoxycorticosterone acetate, 2 mg/day, and cortisone, 20 mg/day,, had normal Na and K balances for the 8-day control period, but showed retention of Na during the 8 days of oestradiol treatment (1 mg/day). Within the 8-day recovery period Na balance was restored to normal. The decrease in Na excretion during oestradiol administration was not accompanied by decreases in the clearance rates of creatinine. Three of the 5 dogs developed hypertension during oestradiol treatment. K balance was not affected by oestradiol administration. Four dogs, maintained only with cortisone 20 mg/day, were in negative Na balance in the 8-day control period, but during the 8 days of oestradiol administration showed retention of both Na and K. Na balance was again negative during the 8-day recovery period. These studies demonstrate that oestradiol produces Na retention independent of an action to increase adrenal steroid secretion.
