Biological properties and genome structure of the Kazakh isolate K1 of Tobacco Mosaic virus

Isolate TMV-K1 was selected from a population of Kazakh strain of tobacco mosaic virus (TMV). Biological properties of TMV-K1 were compared to those of the closest TMV-K2 and tomato mosaic virus (ToMV). It was shown that TMV-K1 was defective with respect to the remote transport; it was found to be more temperature-sensitive and accumulate larger amounts of free envelope proteins in infected plants. The comparison of nucleotide sequences of TMV-K1 genome and TMV-K2 and ToMV genomes revealed 42 nucleotide exchanges; only 11 exchanges led to amino acid exchanges: five in replicase and 6 in envelope protein. It is suggested that mutations in envelope protein determine differences in the infection symptoms and remote transport between TMV-K1, TMV-K2, and ToMV.
