Nine new breakpoints (junction sites) of the BCR and ABL genes in a chimeric oncogene BCR/ABL from translocation t(9;22) were studied in patients with chronic myeloleukemia. It was demonstrated for the first time that some breakpoints in these genes are identical in different patients. DNA fragments containing BCR/ABL breakpoints were amplified using the PCR walking technique. The approach, called reverse primer walk (RPW), is based on a modification of the PCR method with a random annealing of primers.
