Securinine induced apoptosis in human leukemia HL-60 cells

The pharmacological effects of securinine, isolated from Securinega suffruticosa, were investigated in human leukaemia HL-60 cells. Inhibition of proliferation was measured using MTT assay, and the number of apoptotic cells was determined by flow cytometry. DNA fragmentation was visualized by DNA agarose gel electrophoresis and cellular changes were observed under the electron microscope. Securinine (5-80 mg/litre) elicited morphological changes associated with apoptosis and DNA fragmentation in a concentration-dependent manner. It inhibited HL-60 cell proliferation in a time- and concentration-dependent manner (IC50 value of 27 mg/litre after 12 h of treatment).
