Mutagenic and toxic effects of methylisocyanate (MIC) in Salmonella typhimurium

MIC was tested for possible mutagenic and toxic effects in the Ames Salmonella/microsome mutagenicity test both in the presence and absence of S9 mixture (rat liver S9 fraction+cofactors) by using five different tester strains, TA 97a, TA 100, TA 102 and TA 104 of Salmonella typhimurium. MIC did not exhibit any mutagenicity in three tester strains of Salmonella but a weakly mutagenic effect was observed in TA 100 and TA 104 strains in the absence of rat liver S9 mixture. Incubation into S9 mixture reduced the mutagenic activity. At relatively higher concentrations MIC was cytotoxic to all the five strains both in the presence and absence of S9 mixture.
