Teratogenic effects of food addictive ethyl-p-hydroxy benzoate studied in pregnant rats

Experiments with rats were performed to pursue the biological effects of ethyl-phydroxybenzoate (E.B.) in pregnancy. Pregnant and non -pregnant rats of Wistar strain were used. Tests for acute toxicity were performed in three groups with E.B. orally administered at 10c,"'0, 1% and 0.1%. Tests for teratogenicity were made in fetuses from three groups of pregnant rats with E.B. orally given in the critical period at the above concentrations. Fetal external features were observed, and visceral and osseous conditions were examined with Wilson's method and Dawson's method, respectively. Growth conditions of neonates were observed for about one month after their birth while they were breast-fed by mother rats. No noticeable acute toxicity could be identified (200 mg/kg). Nor was there any sign of teratogenicity identifiable in fetuses in different E.B. concentration. From the 10% E.B. group, some fetuses appeared with low body weight, and there were some instances of malformations of bones and viscera attributed to mothers malnutrition. Neonatal growth curves showed no abnormal trends. The administration dose of 0.1% E.B. (3.9 g/kg) in rats, is accounted comparable to 240 g per 60 kg body weight for human adult. These results show E.B. in the tested range of levels do not cause any significant ill effect in rats.
