Demonostration of Mycoplasma hyorhinis and Mycoplasma hyosynoviae in lesions of experimentally infected swine by immunofluorescence

M. hyorhinis and M. hyosynoviae were demonstrated in tissues of experimentally infected pigs by means of the fluorescent-antibody technique. Diethylaminoethyl cellulose chromatography of fluorescein isothiocyanate-conjugated immunoglobulins was better than several other procedures for reducing non-specific fluorescence. Serosal exudate on visceral organs from pigs infected with M. hyorhinis contained cocci and larger, irregularly spherical bodies. Fluorescent coccoid forms were observed in the superficial cell layers of synovial membranes from 9 of 15 arthritic joints of these animals. Fifteen of 41 synovial fluid samples from which m. hyorhinis had been isolated were positive by immunofluorescence. Networks of fluorescent filaments were observed in sections of synovial membranes and lymph nodes from pigs experimentally infected with M. hyosynoviae. Fluorescence was observed mainly in the subsynovial area of the synovial membrane, but small coccal aggregates and filaments were seen irregularly between and on superficial cell layers. The organism was detected in all of 11 synovial membranes and in 14 of 35 synovial fluid specimens of joints from which the organism had been isolated.
