Evidence that the D13S25 locus lies close to a potential tumour suppressor gene implicated in the pathogenesis of B-CLL has been based on detection of LOH and bi-allelic loss using the pH2-42 probe. The Sspl polymorphism detected by this probe has been identified by sequencing adjacent clones and a polymorphic (TA)-n repeat has been found. Amplification of the region encompassing both polymorphic markers by PCR increases the informativity to 80%.
