The mechanisms by which specific dietary fatty acids exert their effects at the genomic level were studied using in vivo (rats) and in vitro breast cancer model systems. The results pointed to the possible role of p53 phosphorylation, which could mediate cell cycle distribution and altered cell growth, although the mechanisms involved in such phosphorylation in normal vs. transformed cells as well as in vivo mammary gland obtained cells are yet to be explored. Furthermore, the multifunctional role of p53 in cell growth is also anticipated to yield additional changes to the cell at the level of transcription and repair. In spite of the inability to identify different frequency or ras mutation in n-6 and n-3 fed rats, it is likely that certain genomic alterations, or their rate, would be selectively modified by specific types of fatty acids as tested in this study.
