Improved detection of the beta-agonist clenbuterol by analysis of retina extracts

Eye and liver samples removed from 4 Hereford heifers experimenta1ly medicated with 10 times (1.6 micro g/kg) the therapeutic con1n of beta -agonist clenbuterol (CBL) for 30 days were screened by enzyme immunoassay for CBL concn; findings were confirmed using gas chromatography mass spectroscopy. Retina to liver CBL concn ratios were 25:1 and 32:1 in 2 of the heifers who were slaughtered before drug withdrawal and 93:1 and 101:1 in 2 heifers who were killed after a 25 day withdrawal period. Immunoassay screening of eye and liver samples taken from 18 highly suspect bovine carcasses at slaughter showed samples of liver from 10 were positive, CBL was confirmed in all 10 by gas chromatography. Retinas from all these animals had higher concn of CBL. Of the 8 liver samples screened negative for CBL, analysis of the corresponding retinas showed that the drug was present in 5 cases. It is suggested that retinal analysis may permit CBL detection in animals where the withdrawal period has reduced the liver concn below the limit decision of the assays. However the small quantity of retinal material available may reduce the suitability of this matrix for routine screening and confirmation analyses.
