A simplified colloidal iron technique for staining ground substance acid mucopolysaccharides was compared with the Rinehart and Abul Haj method in 60 different paraffin embedded tissues. Although the total staining time was only a few minutes using the simplified technique, the selectivity was similar to that of Rinehart's method. Another advantage is the ready availability of dialyzed Fe, which saves time in preparing various working solutions. The proposed technique seems to be quite promising when demonstration of ground substance acid mucopolysaccharides is essential.
