The optimal Na ion concentration in preventing hemolysis of concentrated red cell dilution

The present experiments were carried out for the purpose of detecting the optimal concentration of the Na ion when diluting the concentrated red cell (CRC) isotonically. The CRC was stored in a CPD pack and was divided into three groups (fresh, 1 wk stored, 3 wk stored). The CRC was rinsed before the experiments and mixed with the same volume of different kinds of isotonic solutions. The degree of hemolysis at 2 hrs, 6 hrs and 24 hrs was checked after mixture, and the degree of hemolysis was checked by distilled water and this value was taken as a control. The results of these experiments were expressed by a hemolysis ratio. The relation between the hemolysis and the pH of solution (between 5 to 9) diluted by the glucose and xylitol solutions was also studied. We found that the lower concentration of Na ion resulted in a higher hemolysis ratio. A low hemolysis ratio occurred with both 10% maltose and 5% xylitol. However, a high hemolysis ratio occurred with 5% glucose. No correlation was found between hemolysis ratio and pH of between 5 to 9 of the glucose or xylitol solution.
