A three-month subacute toxicity study of bis (2,3,3,3-tetrachloropropyl) ether was performed on male and female Wistar rats. The test substance was given ad lib. At dose levels of 0 (control), 600, 3000 and 15000 ppm in the diet. All animals in the 15000 ppm group of both sexes became emaciated and died within 30 days of treatment. Slight depression of body weight gain was also observed in the 600 and 3000 ppm groups of the males. The MCV value was slightly decreased in the 600 ppm group of the females and in the 3000 ppm group of both sexes. Increases in phospholipids and total cholesterol in the serum were noted in the 3000 ppm group of both sexes. Triglycerides were also significantly increased in the females. Similar findings were observed in the 600 ppm group of both sexes. Increases in liver weight was observed in the 3000 ppm group of both sexes. Slight increases in liver and kidney weights were also noted in the 600 ppm and 3000 ppm groups. Hepatocellular necrosis or swelling was seen in the 15000 ppm group (dead animals) of both sexes. Slight hepatocellular vacuolization and swelling were also observed in both males and females of the 3000 ppm group. The results suggest that the liver is the target organ for toxicity of bis (2,3,3,3-tetrachloropropyl) ether.
