Advantages of the 2FMC macromolecular medium in the determination of Rh (D) antigenicity

The use of the macromolecular medium 2FMC (Fycoll-Methylcellulose) in microtiter plate for Rh (D) antigen determination was compared with the classical tube and plate techniques. Eleven Rh (D)+ red cell samples distributed into 7 Rh (D)+ and 4 Rh(D)-(Du)+ samples which had been previously tested by tube technique and indirect Coombs' test were titrated with doubly diluted brand reagent antibodies. The results attained with 2FMC showed higher sensitivity than the classical methods, since the former proved capable of detecting all the Rh (D)-(DU)+ samples, making the use of the antiglobulin test unnecessary as opposite to the latter, which require the Coombs's test to achieve similar sensitivity. No false positive results were found with 2FMC in spite of its higher sensitivity.
