Electron microscopic and morphometric studies of the hippocampus of the NZB mouse

By using electron microscopic and morphometric methods, the volume densities of the entire mitochondria, the mitochondria in boutons and the boutons were determined in stratum moleculare of the fascia dentata (Mol Fd) and in substratum eumoleculare of CA1 (Eumol CA1) in the hippocampus of New Zealand Black (NZB) and Carworth Farm Webster (CFW) mice (control animals). Concerning these parameters the study showed that there were small, statistically non-significant differences between the two mouse strains. Accordingly in the NZB mice the volume density of the entire mitochondria was in Mol Fd 4.3% and in Eumol CA1 3.5% and the volume density of the mitochondria in boutons in Mol Fd 3.8% and in Eumol CA1 2.2% lower than in the CFW mice. Whereas the volume density of the boutons in Mol Fd was equal in both mouse strains, in Eumol CA1 of the NZB mice it was 3.5% higher compared with the CFW mice. Comparing the investigated hippocampal layers within each mouse strain, in both strains higher volume densities of boutons (by about 38% in the CFW and 34% in the NZB mice) and of mitochondria in boutons (by about 64% in the CFW and 61% in the NZB mice) were determined in Mol Fd compared with Eumol CA1. The volume density of the entire mitochondria in Mol Fd was in the CFW mice about 9% and in the NZB mice about 10% lower than in Eumol CA1. Concerning the investigated parameters the results of the CFW mouse were well in accordance with other species (rat, guinea pig). Findings in the NZB mouse which is considered as an animal model for cognitive deterioration with aging are discussed in connection with stereological and enzyme-histochemical findings in old rats.
