1) A single intravenous administration of z gram of dl-biotin per kg. produces no toxic signs in mice. Multiple large doses of biotin given intravenously to dogs or per os to mice and normal rats cause no noteworthy signs of toxicity. 2) Relatively high concentrations of biotin do not alter the contractions of the isolated uterus or intestine of the normal rat. 3) Biotin produces no 'significant signs of irritation following local application to the cornea (rabbit), intradermal injection (guinea pigs) or intramuscular injection (rabbit). 4) In the anesthetized normal rat large doses of biotin intravenously produce no effect on blood pressure, respiration, or intestinal circulation. The secretion of free hydrochloric acid by the gastric glands appeared to be depressed by d-biotin. 5) A large single oral dose of d-biotin to normal rats produces no deviation from the normal renal function or metabolic rate. Repeated oral administration of large doses of biotin has no effect on hepatic function, renal function, blood fibrinogen, rectal temperature, sensitivity of nerve endings, skin regeneration or healing of scalded skin in the normal rat.
