To minimize the introduction of artifacts in worm material that is obtained in the field, it should be snap frozen in liquid nitrogen on site, and kept frozen in transit. This should then be plunged into the solubilization buffer of choice with the following protease inhibitors added: PMSF (phenylmethyl-sulfonyl-fluoride) 5 mM final concentration, 1,10 phenanthroline 5 mM, and N-ethylmaleimide 10mM, and rapidly homogenized by mechanical means.
