Selection of non-flagellated and non-piliated mutants from Pseudomonas aeruginosa strain TPB-1

These different mutants, flagellated and non-piliated (F+P-), non-flagellated and piliated (F-P+), and non-flagellated and non-piliated (F-P-) bacteria were selected from Pseudomonas aeruginosa strain TPB-1. By selecting the non-spreading colonies in soft agar, non-motile mutants were obtained from all the mutants occurring spontaneously during a prolonged stationary culture in a liquid medium. Non-flagellated (F-) mutant was selected from the non-motile mutants which agglutinated slightly with anti-flagella serum. Flagellated (F+) colonies were selected from motile colonies which agglutinated strongly with anti-flagella serum. Non-piliated (P-) mutants were then selected from the colonies of F+ and F- organisms which agglutinated slightly in some delay with anti-pili serum, respectively. Piliated (P+) colonies were selected from the colonies of F+ and F- organisms which agglutinated strongly with anti-pili serum, respectively. Presence or absence of flagella and pili was confirmed by electron microscopy.
