Production of high titrating antisera against BVD virus from swine and its use for direct immunofluorescence and immunoperoxidase technics

Antibodies against bovine viral diarrhoea virus (BVDV) raised in pigs by an anamnestic response method were used for the production of FITC and immunoperoxidase conjugates. The whole procedure including immunization of pigs, conjugation with FITC and peroxidase, and the testing of the conjugate is described and discussed. Six weaner pigs were intranasally inoculated with BVDV strain Osloss/2482 and eight weeks after primary inoculation intranasally challenged with swine fever virus strain Alfort. The serum samples collected two weeks later exhibited high homologous and also heterologous neutralizing antibody titres against BVDV strain NADL and swine fever virus strains Alfort and 331. The immunoglobulin fraction of one serum sample with a homologous antibody titre of 1:6000 was coupled to FITC and to horseradish peroxidase, respectively. Both types of conjugates may be applied with advantage in direct techniques on cell cultures (virus isolation, NIF test, direct NPLA) or sections prepared from frozen or paraffin-embedded organ tissues.
