Effects of placental glycoproteins on alloantibody formation, graft versus host reactivity and mixed lymphocyte reaction

Placental proteins from C-57 Bl/Ks (H-2d) mice were fractionated employing (NH4)2SO4 precipitation. Con-A Sepharose affinity chromatography and Sephacryl S-200 sieve chromatography. Various fractions (SF1-SF6) were evaluated for their effect on immune deviation in terms of alloantibody formation in C-57 Bl/Ks mice using A/J (H-2a) spleen lymphocytes as target cells, local graft versus host (GVH) reactivity in (C-57 Bl/Ks .times. A/J)F1 hybrids, and mixed lymphocyte reactivity (using C-57 Bl/Ks lymphocytes as responder cells and A/J lymphocytes as stimulator cells). It was observed that the formation of early Ig isotypes (IgM) is suppressed by glycoprotein fractions, SF1 and F6 and enhanced by a protein preset in SF3/SF4. The popliteal lymph node assay in F1 hybrids showed a significant reduction in GVH stimulation index in presence of SF2 and SF6, representing an average molecular weight of 300 and 40 KD respectively. Though SF6 completely abolished the proliferation of lymphocytes in mixed lymhocyte reaction, SF2 diminished mixed lymphocytes reactivity by 30% (P<0.01). It is concluded that placenta contains glycoproteins which can affect alloantibody formation (suppression and enhancement associated with different fractions) and cellular immune responses against paternal histocompatibility antigens during pregnancy.
