Electron microscopic localization of acid phosphatase in the injured dental pulp

The authors studied the submicroscopic localization of acid phosphatase in the calf dental pulp following injury and direct capping with Calxyd. The reaction of the dental pulp was investigated 7 and 21 days after the capping. Acid phosphatase activity was found to be high in comparison to its level under physiological conditions and was positive in all cell components of the dental pulp. Strong reaction was found in the endothelial cells mainly in lysosomes, Golgi complex, endoplasmic reticulum profiles, and plasmalemmal vesicles. Fibroblasts displayed the same enzyme localization. High acid phosphatase reaction occurred in odontoblasts of the newly-formed barrier. The enzyme was present mainly in the smooth endoplasmic reticulum system and in the Golgi complex. The high level of phospatase activity also persisted for 21 days. The findings and literature are discussed in the paper. The results of the present study demonstrated that in the period when the defect had already morphologically healed the biological processes in the dental pulp remained on a different metabolic level and the tissue had not returned to its physiological limits. This fact must be respected in the treatment of teeth with an injured dental pulp.
