Morphological development of osteoclasts as revealed by monoclonal antibodies

We have already produced monoclonal antibodies against mature chicken osteoclasts1.2. Using these antibodies, osteogenetic process of chick embryo was studied immunohistochemically, and the results were compared with acid phosphatase and esterase stainings. On day 10 of incubation, imunoreactive mononuclear and multinuclear cells first appeared at the erosion site of hypertrophic cartilage. These cells appeared to invade cartilage matrix. On day 12, positive cells were found attached to the hypertrophic cartilage and the calcified cartilage matrix surrounding the developing marrow cavity. Esterase activity, which is reported as a marker for the monocyte/macrophage, was also positive in these immunoreactive mononuclear cells. Reactivity against the monoclonal antibodies was not seen in the mesenchymal tissue until vascular invasion first appeared. These results suggest that osteoclast precursors may have some relation to monocyte/macrophage cell line and emigrate via the blood stream to the region of bone development to become mature osteoclasts.
