A double antibody sandwich ELISA was developed for detecting rotavirus antigen from faeces of calves with diarrhoea, using scopyrol plates. Anti-rotavirus IgG (NCDV strain) from rabbit was used both as capture antibody (1 mu g/ml) and as peroxidase conjugate (1:20 000). ELISA reaction was evaluated by parallel application of pre-immune IgG. Sixteen of 72 faecal samples were positive to ELISA and counter-immuno-electroosmophoresis (CIEOP), and five samples were positive to ELISA only. Average virus shedding amounted to 4.6 days according to ELISA and 3.2 days by CIEOP. The antigen titres of one virus-positive faecal sample were 1:128 by CIEOP and 1:4000 by ELISA. Cell culture fluids containing corona, herpes and paramyxoviruses responded negatively to both ELISA and CIEOP.
