Chromosome studies in 20 cases of myelodysplastic syndrome

According to the criteria of FAB in 1982, the chromosomes of 20 patients with myelodysplastic syndrome (MDS) were studied and compared with 15 patients with non-malignant hematological disorders. The chromosomal preparations were made from bone marrow aspirates directly without in vitro culture. The slides were processed by the trypsin-Giemsa banding technique. Karyotypes were described according to the International System for Human Cytogenetic Nomenclature. It was found that the aneuploid was 33.3% in MDS group, while the aneuploid in the control group was 14.8%. The difference was statistically significant. The chromosomally structural aberrations in MDS group was higher than those in the control group. The major type of chromosomal aberrations were chromosome break and chromatid break. Of the 20 patients, seven had a chromosomally abnormal clone. The chromosome aberrations included +8, -21, 7q-, -Y, etc. Two of the 7 patients with abnormal karyotypes evolved into acute leukemia (AL); one of the 13 patients with normal chromosome transformed into AL. The present study supports that MDS have nonrandom abnormalities of chromosome, and increasing incidence of aneuploid. Part of MDS have the quality of malignant tumor. We have not reached a conclusion of the relation between the karyotype and the prognosis as yet, because the period of observation was too short.
