The feasibility of analysing lymphocyte subsets in cytocentrifuge preparations using a simple immuno-peroxidase staining technique is assessed. The method correlates extremely well with classical immuno-fluorescence of lymphocytes in suspension, offering a reliable alternative for the determination of lymphocyte sub-populations in both fresh and cultured cell samples. Antibody labelling procedures are carried out entirely on fixed cytocentrifuge preparations, which may be stored for several weeks prior to immuno-staining without loss of activity.
