Determination of metabolic activity in bone tissue in rats using 24-hour whole-body retention of 99mTc-diphosphonate

The 24-hour whole-body retention of 99mTc-diphosphonate is an index of the level of metabolic turnover of the bone tissue (2). In addition to a diagnostic use (5) the authors have introduced a similar experimental method with a whole-body measurement of general-anaesthetized rats in a large-volume scintillation detector (first measurement 10-15 minutes after the injection of 99mTc-methylenediphosphonate Amersham, England, or 99mTc-hydroxyethylidenedisphonate Rotop, GDR, the second measurement after 24 hours.sbd.expressed as a percentage of the frequency of the first measurement). The mean value .+-. sx in the adult female rats proved to be 34.0 .+-. 0.9% for 99mTc-MDP and 24.1 .+-. 0.3% for 99mTc--HEDP. The authors verified some methodical questions and came to the following conclusions: a) Retention is not influenced by the amount of disphosphonate at the dose in the range of 0.0005 mg.sbd.0.005 mg.sbd.0.05 mg; b) The comparison revealed that in addition to the i.v. injection of the labelled preparation it is also possible to use intraperitoneal administration. c) The percentage retention is not influenced by considerably different body mass caused mainly by variable content of fatty tissue and differentiated food supply. The determination of 24-hour whole-body retention of 99mTc-disphosphonate in the presented way appears to be a method suitable for the observation of metabolic activity of bone tissue in model conditions in the experimental animals.
