Changes of glutathione status and criteria of mitochondrial respiration during normoxic perfusion and ischemic storage of rat liver

The oxidative stress and functional and structural criteria of mitochondria were studied on rat liver during ischemic storage or normoxic perfusion at 5 and 25.degree. C, respectively. The release of glutathione disulfide as a sensitive indicator of metabolism of reduced oxygen species increased with temperature and was lower in perfusion compared with ischemic storage. The respiratory control ratio decreased owing to diminution of state 3 respiration; there was no sign of uncoupling of oxidative phosphorylation. There was found to be a linear correlation between the inhibition of respiration by carboxyatracyloside and the rate of GSSG efflux. At 25.degree. C there were observed structural alterations in mitochondria which are more distinctly marked in ischemic storage. These findings were discussed with respect to the role of reduced species of oxygen in ischemia and organ preservation.
