New chromosome banding techniques with base specific antibiotics and fluorochromes for qualified identification of marker chromosomes in leukemia and related disorders

Demonstration of sequential R-, C-, and Q-bandings on the same metaphase plate was successfully performed using anticancer antibiotics and newly developed fluorochromes- actinomycin D, chromomycin A3 specific for guanine-cytosine(G-C) rich DNA, and distamycin A and DAPI specific for adenine-cytosine(A-T) rich DNA. A dual karyotype consisting of G- and C-bands was also demonstrated on the same metaphase plate. Using these methods, chromosome constitutions of nine cell lines established from leukemias and lymphomas, and thirty-five patients with leukemia and related disorders were examined. Complicated structural rearrangements could precisely be analyzed with these techniques, most of which posed difficulties when conventional banding techniques were used singly. These techniques save time, and are excellent tools for studying the cytogenetics of hematologic malignancies and solid tumors, especially analyzing marker chromosomes.
