Involvement of eosinophils in leukemic clones: cytogenetic study in eosinophilic colonies from acute myelogenous leukemia associated with 8;21 translocation

Cytogenetic analysis was performed in the eosinophilic colonies grown in the marrow culture from a patient with acute myelogenous leukemia (FAB classification, M2) associated with 8;21 translocation. The marrow, in which 50.2% myeloblasts and 5.6% eosinophils were observed, gave rise to leukemic colonies and eosinophilic colonies identified by Luxol-Fast-Blue staining. The cytogenetic studies were performed in individual colonies and in pooled colonies. The experiments were done on two occasions. The first study showed 8;21 translocation in 15 out of 16 metaphases in randomly selected colonies from the dishes in which 65% of the total colonies were leukemic colonies and 35% were eosinophilic colonies. The second study showed t(Cq-; Gq +), considered to be 8;21 translocation, in all metaphases (6/6) in selectively chosen eosinophilic colonies. The granulocyte-macrophage colonies and erythroid burst colonies obtained from the marrow in remission revealed normal karyotype. Our present studies on this case suggest the involvement of the eosinophilic series in leukemic clone.
