Pharmacokinetics of nifedipine and prediction of plasma concentration on essential hypertensive patients

The pharmacokinetics after oral administration of nifedipine was studied in 10 healthy male volunteers. The plasma concentration-time course pattern of nifedipine was quite liable to variation within individual subjects and a great variation of absorption rate constant was observed from the results of pharmacokinetic analysis. A program which could predict the blood level curve after repetitive dosing of various dose sizes at non-uniform dosing intervals was developed; the prediction of plasma levels after multiple dosing of nifedipine was attempted on 3 essential hypertensive patients by using this program. The predicted plasma levels were in good agreement with observed plasma levels; therefore, this program is suited to designing optimum dosing schedules for individual patients. A significant correlation between the plasma concentration of nifedipine and the change in systolic and diastolic blood pressure reduction was observed.
