"in vitro" growth acceleration of CFU-GM proliferation: a new criterion of measure of proliferation features of granulocytic progenitor cells

We studied bone marrow granlocytic progenitor cell growth behaviour in agar culture in 18 healthy volunteers, by scoring dishes at two different sequential times, on the 7th and on the 14th day, in order to estimate the dynamic events of proliferation sustained by CFU-GM subtypes that concur to maintain the dynamic equilibrium of granulopoiesis. We propose an evaluation criterion that is based on the biologic parameter (growth acceleration) for cluster-and colony-forming populations in the two incubation intervals. Growth accelerations are represented by vectors, having as (magnitudes) the growth on the 7th day or the percentage variations on the 14th day in relation to the 7th; (directions) corresponding to the abscissa or ordinate according to the scoring time and (senses) positive or negative according to increase or decrease of growth. Adding these vectors we obtain resulting vectors or development phenomena for clusters and colonies respectively, throughout the entire culture period. By transforming Cartesian coordinates into polar coordinates, vertices distribution of resulting vectors lies in two polar areas A and B; they show different growth behaviour in agar of cluster- and colony-forming CFU-GM. By extrapolation we obtain a third area C, that would show the range of oscillation within which autoregulation of dynamic equilibrium of granulopoiesis occurs.
