Effect of platelet-derived growth factor on the in vitro cell proliferation of chick embryo skin

Mitogenic factors of platelet origin apparently can act upon fibroblasts, smooth muscle cells and neuroglial cells. On this basis, the possibility of a differential effect in the multiplication of chicken embryo skin tissue in vitro by human serum obtained, respectively, from platelet rich plasma (SPRP) and platelet poor plasma (SPPP) was examined. With this aim, organotypic cultures of the lower limbs of the chicken embyro at the 6th day of incubation were prepared and cultivated in vitro with SPRP or SPPP in the presence of 3H-labeled thymidine for 2-4-7 days. Parallel cultures were prepared without thymidine for light microscope observation. Morphologic results demonstrate normal evolution of cell multiplication and differentiation of the skin with both types of serum. 3H-labeled thymidine incorporation demonstrated similar values. This gives a starting point for further study, for example, in the field of vascular pathology using human skin tissue in culture.
