Chromosome mapping by in situ hybridization of cloned human DNA sequences

"In situ" hybridization allows the chromosomal localization of cloned DNA sequences. In the present paper we report "in situ" hybridization experiments to localize: (A) 7SL RNA genes, (B) the sequences homologous to a clone (29 .times. 3) from a .times. chromosome library and (C) the human thyroglobulin gene. (A) "In situ" hybridization with the SAU 3A fragment of a 7SL RNA gene shows extensive labeling in different chromosomes, indicating that the 7SL genes (approximately 1000 copies) are spread in the human genome. (B) The DNA sequences 29 .times. 3 (approximately 150 copies in the genome) significatively hybridize with the chromosomal sites: X q22-25, 4 p15-14, 5 q31-33 e 16 q11-13. (C) the localization of the autoradiographic grains, obtained with the 2 Kilobases thyroglobulin genomic probe, suggests a sub-regional assignment of the human thyroglobulin gene to 8 q23-24.
