Evaluation of colorimetric method using thiobarbituric acid for the determination of glycosylated hemoglobin

It was examined whether 5-hydroxymethyl furfural (5-HMF) is the only substance obtained by hydrolysis of the products that have undergone Amdori rearrangement and by the degradation process of the resultant Amadori product. In the colorimetric TBA method for the determination of glycosylated Hb, the measurement using the absorbance of 5-HMF had some problems, i.e., acid-hydrolysis products of HbA1 and glycosylated protein (or albumin) contain substances other than 5-HMF, and 5-HMF is not an end product of glycosylated Hb. Thus strict conditions for this method should be established in the future.
