Typing of herpes simplex virus isolates by different serologic & biologic methods

Three serologic techniques, i.e., microneutralization (MN), inhibition of indirect hemagglutination (IHAI) and immuno-fluorescence (IF), and 3 biologic parameters.sbd.growth characteristics in monolayers of chick embryo fibroblasts (CEF), characteristics of cytopathic effect on vero cells (CPE) and characteristics of pocks on chorioallantoic membrane of embryonated hen's egg (CAM), were evaluated for their efficacy in typing fresh isolates of herpes simplex virus (HSV). A complete correlation was observed among different tests in typing oral isolates of HSV-strains. In typing genital isolates, CPE on vero cells gave the most variable results. Comparison of the remaining 2 biologic parameters with the 3 serologic tests showed concordant results in 14 out of 18 virus isolates and discordant results in 4 HSV-isolates. In addition, it was observed that all the HSV-isolates from oral lesions and 11 out of 18 isolates from the genital lesions were typed as HSV-1. Only 3 isolates (16.7%) from acute genital lesions were HSV-2.
