Effect of centpropazine on hepatic drug metabolizing enzymes in rat

Centpropazine a new antidepressant compound developed by the Central Drug Research Institute, Lucknow [India], was subjected to studies for its effect on hepatic microsomal aminopyrine N-demethylase, aniline hydroxylase and electron transport components, cytochrome P450, cytochrome b5 and heme in male albino rats for a period of 7 days. Although there was no significant alterations in aminopyrine N-demethylase activity, aniline hydroxylase activity, cytochrome b5 and heme contents showed continuous decrease, and cytochrome P450 increased up to the 3rd day and subsequently decreased up to 7th day of drug treatment. The change in microsomal protein content tallied with the alteration in cytochrome P450 concentration, while the sum of cytochrome P450 and cytochrome b5 was equivalent to the heme content.
