The degranulation of rat peritoneal mast cells by the compound 48/80 was studied either on cells collected by peritoneal washing or on peritoneal fragments. During degranulation, the membrane folds became smaller. The underlying granules plainly deformed the membrane which subsequently opened at the point of deformation. Then the granules were extruded. In the mast cells marginating on the peritoneal fragments, the membrane modifications were more unobtrusive than those of cells collected by peritoneal washing.
