Intercellular bonding in the erythroblastic nest (EN) is reversible and dependent on divalent cations

Dispersing erythroblastic nests (EN) is the initial step in separating the central cells from the erythroblasts. The effects of 3 reagents: EDTA, ethyleneglycol-bis-(.beta.-aminoethyl ether)-N,N'-tetraacetic acid (EGTA), and citrate-phosphate-dextrose (CPD) were studied to develop a dispersal procedure and to gain some insight into the nature of the intercellular bonding in these erythropoietic tissue structures. CPD, EDTA and EGTA all dispersed EN. CPD was the most effective because cell viability was maintained and cell reassociation did not occur. When cation chelators were added, the bonds between the dispersed central cells and erythroblasts were seen to be reversible Ca2+ or Mg2+ dependent.
