Influence of non-steroidal anti-inflammatory agents on tolbutamide hypoglycaemia

Drug interaction between tolbutamide and other commonly used nonsteroidal antiinflammatory agents was investigated. The subjects included non-diabetic rabbits and healthy human volunteers. Cross over design was employed in the animal as well as clinical study. Tolbutamide caused a decline in blood sugar by 22-23% from the fasting value both in animals and human volunteers. None of the antiinflammatory agents used caused, by themselves, any significant change in blood sugar level. When given prior to tolbutamide, indomethacin, ibuprofen and N-.beta.-phenylethyl anthranilic acid significantly (P < 0.001) antagonized tolbutamide hypoglycemia while phenylbetazone did not modify the later. The relevance of such interactions in the therapy of diabetics in clinical practice was highlighted.
