Activity of two polyene and two imidazole antimicrobics on Candida albicans in human fibrin clots

The vegetations of infective fungal endocarditis were simulated by preparing human fibrin clots containing 105 CFU of C. albicans per clot. Clots were incubated in human serum without drugs or with equimolar conc. (2.5-20 nmol/ml) of amphotericin B, miconazole, guanido derivative of amphotericin B (DAB) or ketoconazole. Over a period of 4 days of incubation, clots were resuspended in freshly prepared serum-antifungal agent every 12 h. Neither imidazole at any conc. tested significantly prevented growth of C. albicans. Both polyenes reduced the CFU/clot with amphotericin B significantly more active than DAB.
