Effects of nitrous oxide on human cell lines

Hematopoietic and nonhematopoietic cell lines [chronic myelogenous leukemia K562 cells, promyelocytic leukemia HL-60 cells, T-cell acute lymphocytic leukemia TALL cells, Raji Burkitts lymphoma cells, RPMI 1788 B cells, NCTC 2544 fibroblasts, HMV 143 melanoma cells and Okajima stomach cancer cells] cultured in media containing either 5-methyltetrahydrofolate or 5-formyltetrahydrofolate grow well to the same extent. However, when these same cell lines are grown in the presence of nitrous oxide, selective growth inhibition can be shown for hematopoietic cells cultured in 5-methyltetrahydrofolate-containing media. These cells also demonstrated a decreased ability to suppress [3H]-thymidine incorporation into DNA in deoxyuridine suppression tests. [This finding is related to earlier reports of an antileukemic effect of nitrous oxide.].
