Effect of maternal hypervitaminosis A during late gestation on pre- & postnatal lung maturation in rat

On days 16 to 18 or 17 to 19 of pregnancy CF rats were given 15 000 or 20 000 IU vitamin A by mouth. Mean volume and weight of amniotic fluid were significantly greater for foetuses of rats given vitamin A than for those of controls. Histological study showed that lungs of foetuses of rats given vitamin A had differentiation delayed by 2 days. When treated rats were allowed to proceed to delivery, their offspring showed higher perinatal mortality, smaller litter size and lower birthweight that those of controls; sucking was delayed and they showed respiratory distress. Lungs of offspring 1 to 7 days old showed poor differentiation of pulmonary tissue, thick septae, near absence of secondary septae and delayed vascularization.
