Prevention of certain biochemical changes in acute ischaemic rabbit heart by verapamil

The effect of verapamil on some biochemical changes produced by acute ischemia of rabbit heart was studied. Ischemia was produced by the ligation of the anterior descending branch of the left coronary artery. Verapamil 50-200 .mu.g i.v. reduced the ischemia-induced depletion of glycogen stores and the inhibition of ATPase and succinate dehydrogenase. It lowered the ischemia-induced enhancement in the activity of lysosomal enzymes. The rapid fall in blood pH, blood pressure and heart rate were also prevented. Evidently, verapamil exerts a protective effect over the ischemic myocarium which may be due to its ability to enhance the energy production and lowering of lysosomal fragility. The former effect seems more marked.
