Localization of creatine kinase in developing skeletal muscle of rat

The intracellular distribution of creatine kinase (CK) in developing skeletal muscles of rats was examined by means of differential centrifugation and ultracytochemistry. In the muscles of early postnatal stage, the proportion of CK activity in particle to soluble fraction was higher than that of adult stage. As the skeletal muscle developed after 1 mo., the activity of this enzyme in the soluble fraction rapidly increased against a decrease in that of particle fraction. Electrophoretic pattern of the homogenate showed MB-CK as well as MM-CK in early developmental muscles. Using histo- and ultracytochemical techniques, membrane bound activity could be demonstrated in the sarcoplasmic reticulum and mitochondria. Evidently, CK is predominantly localized in the particle fractions of early developmental muscles, and these muscles depend on different metabolic pathways in obtaining the supplies of energy for muscle contraction and differentiation.
