Fluorometric estimation of taurine in tissue extracts and biological fluids

A fluorometric technique was used for determination of taurine in extracts of rat brain and liver, in human urine and in cows', buffaloes' and human milk. Tissue samples were extracted with 75% alcohol and treated with 6 M HCl to hydrolyse glycerophosphoryl ethanolamine, and milk samples were treated with 5% trichloroacetic acid, before chromatography on Dowex-AG-50W x 8 resin. Interference from cysteic acid was then eliminated by chromatography on a Bio Rad-AG 1 x 8 column. Effluents were treated with fluorescamine, and fluorescence was measured. Taurine concn. in cows' milk, buffaloes' milk and human milk were 4.14 +or- 0.98, 5.90 +or- 0.90 and 33.98 +or- 8.39 mol/100 ml, resp. Recovery of taurine added at 5-60 nM to brain extracts and urine was 96.3-99.5%. Results obtained using this method were in good agreement with those from an amino acid analyser.
