Interaction of 2 membrane active polypeptides, .**GRAPHIC**. and .**GRAPHIC**. from Central Asian cobra venom with rat liver mitochondrial membrane was studied. These cytotoxins did not affect the oxidative phosphorylation of intact mitochondria. Their effects and differences in their action became evident when mitochondria were incubated with the cytotoxins in the absence of oxidative substrate, succinate. While .**GRAPHIC**. acted as an uncoupler .**GRAPHIC**. caused respiratory inhibition without showing uncoupling activity. The 2 toxins affected the mitochondrial polyenzyme systems in an opposite manner. Effect of Ca ions on the action of polypeptides depended on the enzyme system and the venom component. Structural differences in the cytotoxins had a direct bearing on their interaction with mitochondrial membrane.
